Cuproptosis is a form of regulated cell death that is fundamentally different from other known types, such as apoptosis, ferroptosis, necroptosis, or pyroptosis. It is triggered by excessive intracellular copper accumulation and is uniquely dependent on mitochondrial respiration.
It may be triggered by copper overload from either direct copper ion treatment or copper ionophores that shuttle Cu into cells. Copper binds directly to lipoylated proteins in the tricarboxylic acid (TCA) cycle, causing abnormal protein aggregation and proteotoxic stress. This results in the downregulation of iron-sulfur (Fe-S) cluster proteins and a loss of key metabolic enzymes. Cuproptosis only occurs in cells that rely on mitochondrial oxidative phosphorylation.
