Vascular endothelial growth factors (VEGF-A, -B, -C, -D and PGF) are the main regulators of blood and lymphatic vessel growth. VEGF-A binds the receptor tyrosine kinases VEGFR1 (FLT1) and VEGFR2 (KDR) on endothelial cells, with neuropilin coreceptors, while VEGF-C and VEGF-D signal through VEGFR3 (FLT4) to drive lymphangiogenesis. VEGFR2 activates PLC-gamma, PKC, RAS-MAPK, PI3K-AKT and eNOS to promote endothelial cell proliferation, migration, survival and vascular permeability. VEGF expression is induced by hypoxia through HIF1A, and VEGF signaling is a major target of anti-angiogenic therapy in cancer and eye disease.
