T cell receptor (TCR) signaling begins when the TCR recognizes a peptide presented by MHC on an antigen presenting cell, together with the CD4 or CD8 coreceptor. The kinase LCK phosphorylates ITAMs in the CD3 complex, recruiting and activating ZAP70, which phosphorylates the adaptors LAT and SLP76 (LCP2). This activates PLC-gamma1, calcium/calcineurin/NFAT, PKC-theta/NF-kappaB and RAS-MAPK/AP-1 pathways, while CD28 costimulation activates PI3K-AKT. These signals lead to IL-2 production, T cell proliferation and differentiation into effector cells.
