Transforming growth factor beta (TGF-beta) is a multifunctional cytokine that regulates cell growth, differentiation, extracellular matrix production, immune suppression and wound healing. TGF-beta binds the type II receptor TGFBR2, which recruits and phosphorylates the type I receptor TGFBR1 (ALK5). TGFBR1 then phosphorylates SMAD2 and SMAD3, which combine with SMAD4 and regulate target genes in the nucleus. TGF-beta also signals through non-SMAD pathways such as MAPK, PI3K-AKT and Rho GTPases. TGF-beta inhibits growth of normal epithelial cells but promotes fibrosis, EMT and metastasis in advanced cancer.
