Oncogenes are mutated or overexpressed versions of normal genes (proto-oncogenes) that drive uncontrolled cell growth and cancer. Proto-oncogenes include growth factors, receptor tyrosine kinases (EGFR, ERBB2, MET), signaling proteins (KRAS, BRAF, PIK3CA, SRC, ABL1), transcription factors (MYC, JUN, FOS) and regulators of the cell cycle and apoptosis (CCND1, BCL2). They can be activated by point mutations, gene amplification or chromosomal translocations. Oncogene expression profiling is used to understand tumor biology and to identify targets for cancer therapy.
