Interleukin 17 (IL-17) cytokines, especially IL-17A and IL-17F, are produced mainly by Th17 cells, gamma-delta T cells and innate lymphoid cells. IL-17 binds the IL17RA/IL17RC receptor complex and signals through the adaptor ACT1 (TRAF3IP2) and TRAF6 to activate NF-kappaB, MAPK and C/EBP pathways. This induces pro-inflammatory cytokines (IL6), chemokines (CXCL1, CXCL8, CCL20), antimicrobial peptides and matrix metalloproteinases. IL-17 signaling protects against bacterial and fungal infections but also drives autoimmune diseases such as psoriasis, psoriatic arthritis and ankylosing spondylitis.
