Hedgehog (Hh) signaling is a key developmental pathway that controls tissue patterning, stem cell maintenance and tissue repair. In the absence of ligand, the receptor Patched (PTCH1) inhibits Smoothened (SMO), and GLI transcription factors are processed into repressors. Binding of Sonic (SHH), Indian (IHH) or Desert (DHH) hedgehog to PTCH1 releases SMO, which signals through the primary cilium to activate GLI1, GLI2 and GLI3 and induce target genes. Abnormal Hedgehog signaling causes birth defects and drives cancers such as basal cell carcinoma and medulloblastoma.
