Ligand binding activates heterotrimeric G proteins: Gs stimulates and Gi inhibits adenylyl cyclase and cAMP production, Gq activates phospholipase C to generate IP3, DAG and calcium signals, and G12/13 activates Rho GTPases. Signaling is ended by GPCR kinases (GRKs) and beta-arrestins, which also start their own signaling. GPCRs are the targets of roughly one third of approved drugs.
