Focal adhesions are multiprotein complexes that link the actin cytoskeleton to the extracellular matrix (ECM) through integrin receptors. When integrins bind ECM proteins, they cluster and recruit structural proteins such as talin, vinculin and paxillin and signaling proteins such as focal adhesion kinase (FAK/PTK2), SRC and ILK. Focal adhesion signaling activates the PI3K-AKT, MAPK and Rho GTPase pathways to control cell migration, proliferation and survival, and plays an important role in wound healing and cancer invasion.
