Fatty acid and lipid metabolism provides energy, membrane building blocks and signaling molecules. Fatty acids are synthesized by ACACA and FASN, stored as triglycerides, and broken down by lipases and mitochondrial beta-oxidation through CPT1, acyl-CoA dehydrogenases and related enzymes. Cholesterol and other lipids are made through the mevalonate pathway. These processes are controlled by transcription factors such as SREBPs and PPARs and by AMPK, and their dysregulation is central to obesity, diabetes, fatty liver disease, atherosclerosis and cancer.
